首先for a  long time是一个时间段，表示一个持续性的动作，用have been doing比较好
其次something expected一般是表示被别人期望或者预期的
而这里你想表达自己的希望

He was expected to be a doctor. 他曾被期望成为一个医生。（可能是他爸妈对他的期望，但是事实呢？可能他做了别的工作）

前面一句的that或which不能省略，因为他们在从句中是主语。后面一句的that可以省略，因为他是宾语。

想要简单的说的话： Having expected the chance for a long time, I almost lost it.

我认为第一个问题是分词做伴随状语，应该不是介词吧。第二个问题同意ls的意见I almost missed an opportunity that I had been expecting for a long time.:)
